We focus mainly on three pathologies: (i) chronic migraine as a paradigm of chronic inflammatory pain that, in addition, shows a strong sexual dimorphism; (ii) chemotherapy-induced peripheral neuropathy as a model of neuropathic pain syndrome; and, (iii) psoriatic pruritus as a model of chronic pruritus. We aim to validate therapeutic targets to identify and develop drug candidates that restore neuronal sensitivity. Nos centramos principalmente en tres patolog\u00edas: (i) la migra\u00f1a cr\u00f3nica como paradigma del dolor inflamatorio cr\u00f3nico que, adem\u00e1s, muestra un fuerte dimorfismo sexual; (ii) la neuropat\u00eda perif\u00e9rica inducida por quimioterapia como modelo de s\u00edndrome de dolor neurop\u00e1tico; y, (iii) el prurito psori\u00e1sico como modelo de prurito cr\u00f3nico. Pretendemos validar dianas terap\u00e9uticas para identificar y desarrollar candidatos a f\u00e1rmacos que restauren la sensibilidad neuronal. Esta subl\u00ednea se centra en comprender los mecanismos que subyacen a la sensibilizaci\u00f3n proalg\u00e9sica de las neuronas sensoriales, as\u00ed como su desensibilizaci\u00f3n tras la resoluci\u00f3n de lesiones o enfermedades. Nuestra hip\u00f3tesis considera que la cronificaci\u00f3n resulta de una falta o resoluci\u00f3n defectuosa de la sensibilizaci\u00f3n neuronal. Estamos enfocados en tres patolog\u00edas: (i) migra\u00f1a cr\u00f3nica como paradigma de dolor inflamatorio cr\u00f3nico que adicionalmente muestra un fuerte dimorfismo sexual; (ii) neuropat\u00eda perif\u00e9rica inducida por quimioterapia como modelo de s\u00edndrome de dolor neurop\u00e1tico; y (iii) prurito psori\u00e1sico como modelo de prurito cr\u00f3nico. El aspecto com\u00fan de estas tres condiciones es la participaci\u00f3n del sistema sensorial perif\u00e9rico que se sensibiliza al aumentar su actividad electrog\u00e9nica. Nos centramos en el papel de los canales i\u00f3nicos implicados en la generaci\u00f3n de potenciales de acci\u00f3n y en su propagaci\u00f3n, es decir, canales termoTRP, canales Na, Kv y HCN. Adem\u00e1s, investigamos c\u00f3mo la actividad de estos canales se ve afectada por los agentes proalg\u00e9sicos. El objetivo de estos estudios es validar dianas terap\u00e9uticas que se utilizan posteriormente en nuestro programa de descubrimiento de f\u00e1rmacos para identificar y desarrollar f\u00e1rmacos candidatos que restauren la actividad del canal y la sensibilidad neuronal.<\/p><p>[\/et_pb_accordion_item][et_pb_accordion_item title=\"Dise\u00f1o de mol\u00e9culas bioactivas. \" _builder_version=\"4.2.2\" background_color=\"#ab1a2d\" background_enable_color=\"on\" open=\"off\"]<\/p><p>Descubrimiento de f\u00e1rmacos candidatos para la terapia nociceptiva de precisi\u00f3n. La identificaci\u00f3n y dise\u00f1o de mol\u00e9culas bioactivas para diferentes aplicaciones (antiinflamatorias, analg\u00e9sicas y antiprurito) se basa en primer lugar en una estrategia computacional que utiliza modelado molecular, acoplamiento y din\u00e1mica sobre las dianas terap\u00e9uticas validadas. Adem\u00e1s, el cribado <em>in silico<\/em> tambi\u00e9n se aplica a bibliotecas virtuales compuestas por miles a millones de mol\u00e9culas de fuentes naturales y sint\u00e9ticas. Los compuestos de impacto se validan en ensayos de HTS y los compuestos l\u00edder se caracterizan farmacol\u00f3gicamente <em>in vitro<\/em> e <em>in vivo<\/em>. Los candidatos a f\u00e1rmacos seleccionados se licencian a empresas de biotecnolog\u00eda para el desarrollo cl\u00ednico y
