IMAGING ACQUISITION AND ANALYSIS

Microscopio Confocal LSM900 con Airyscan 2

LSM900 with Airyscan 2 confocal microscope shows excellent features in order to acquire high resolution and quality images. Signal to noise ratio is 4 to 8 times increased and super-resolution reaches up to 120 nm.
Airyscan 2 is an area detector with an array of 32 detection elements concentrically grouped. This configuration allows the pinhole to remain open leading to the arrival of more photons and therefore increasing the efficiency. In addition, due to Airyscan 2 Multiplex mode, the acquisition speed raise significantly since with super-resolution up to four pixels of the image are scanned at the same time.
The system is equipped with a motorized high resolution XY-stage and a motorized Z-focus with 10 nm pass. Also, a motorized filter wheel with GFP, Cy3, DAPI and Cy5 filters, several 10x, 20x, 40x and 63x lenses, a LED fluorescent lighting containing 5 modules for RGB-UV, and solid-state diode laser lines of 405 nm, 488 nm, 561 nm and 640 nm are available.
Both, confocal and scanning electronic microscopes from IDiBE, can share the same kind of sample holder and use the same software. Therefore, correlative microscopy CLEM (Correlative Ligth-Electron Microscopy), can be performed in a simple way overlaying and merging information obtained by the two techniques in the same area of the sample.

This equipment has been acquired thanks to an intervention co-financiated by the European Union through the Programa Operativo del Fondo Europeo de Desarrollo Regional (FEDER) and Ministerio de Ciencia e Innovación (Agencia Estatal de Investigación) (PROJECT: EQC2019-005842P).
